چکیده

1. Freezing: the product to be dried is put over shelves in a chamber and its temperature is lowered so that the water (or the other solvent) is frozen. Unfortunately, not all the solvent freezes forming ice crystals, but a certain amount (sometimes significant) remains bonded to the product and must be finally desorbed. In some cases the product (the drug and the excipients) does not crystallize, but forms an amorphous glass which can retain a high amount of moisture. 2. Primary drying: the pressure of the chamber where the product is placed is reduced to a low value, thus causing ice sublimation. This process is carried out at very low temperatures (typically in the range -40°C to -10°C), but it requires energy that is generally supplied through the shelves. 3. Secondary drying: the amount of residual water, strongly bonded to the partially dried product, is reduced to a low value by using high vacuum and moderate temperatures, e.g., 20°C to 40°C. This step is required to ensure the long term preservation of the product.

Demonstrating Functional Equivalence of Pilot and Production Scale Freeze-Drying of BCG

Process analytical technology (PAT)-tools were used to monitor freeze-drying of Bacille Calmette-Guérin (BCG) at pilot and production scale. Among the evaluated PAT-tools, there is the novel use of the vacuum valve open/close frequency for determining the endpoint of primary drying at production scale. The duration of primary drying, the BCG survival rate, and the residual moisture content (RMC...

Modeling and Optimization of Ultrasound-Assisted Osmotic Dehydration with Finished Freeze Drying on Black Cherries – The Effect on Antioxidant Activities

ABSTRACT: In this study, response surface methodology was used to optimize ultrasound osmotic pretreatment with finished freeze drying of black cherries. Freeze drying is a separation process based on the  sublimation phenomenon. This process has the following advantages as compared to the conventional drying process, maintenance of the structure, moisture removal at low temperature (reduced tr...

In-line and Off-line Optimization of Freeze-drying Cycles for Pharmaceutical Products

Model based tools allow optimizing the freeze-drying of a pharmaceutical formulation in a specific freeze-dryer, thus minimizing the duration of the process, beside maintaining product temperature below a limit value to preserve product quality. This paper is focused on the comparison of the results that can be obtained optimizing the process either off-line, using the design space of the formu...

Producion of Nanoparticle Assemblies by Electro-Spraying and Freeze-Drying of Colloids: A New Method to Resolve Handling Problem of Nanoparticles

To resolve handling problem of nanoparticles, due to their small size, a new methodology of electro-spraying and freeze-drying was developed for colloidal nanoparticles of silica and titania to transform them to solid macro-scale nanoparticle assemblies.  The assemblies were then redispersed in an aqueous system to investigate the effect of formulation of original solutions and the process ...

Sperm freeze-drying; effects on sperm parameters

Today, sperm cryopreservation is a routine practice in infertility clinics. One of the alternative methods to preserve sperm is freeze-drying. This process consists of three steps. The first step is sample freezing, during which the solvent is separated from the solvent. Water forms ice crystals and the solute is placed between the ice crystals. Freezing may be done in a separate device or in a...
